Background

The role of alpha-2-antiplasmin (a-2-AP) is in the regulation of intravascular fibrinolysis and functions as the primary circulating inhibitor of plasmin. 1,2 Low levels of a-2-AP is thought to increase the capacity of the fibrinolytic function and may also serve to be beneficial in the treatment of thrombotic disease, acute pulmonary embolism and hepatic repair.

Assay Principle

This is an ELISA for the quantitative analysis of active a-2-AP levels in biological fluid for mouse. This test kit operates on the basis of sandwich ELISA where free, active, a-2-AP complexes with plasmin and is quantified with the use of an HRP labeled secondary antibody.

First the active a-2-AP present in the standard or unknown, complexes with plasmin coated on the microplate. Inactive or complexed a-2-AP is removed in a subsequent wash step. A primary antibody specific for a-2-AP is then added to each well followed by the HRP conjugated secondary antibody. The bound conjugated secondary antibody is detected by the addition of substrate, which generates an optimal color after 10 minutes. Quantitative test results may be obtained by the measure and comparison of the sample and standard absorbance readings when read with a microplate reader at 450 nm.
